Polymorphic HLA-A2-restricted epitopes restricted by HLA-B57 have been described in EBV-transformed lymphoblastoid cell lines and in patients with acute and chronic active EBV infection. These include epitopes restricted by A*0201, A*0205, and A*0206. It has been suggested that HLA-B57 may predispose individuals to develop EBV-associated disorders. To identify other HLA-B57-restricted epitopes of EBV, we have screened a series of recombinant viruses, encoding EBNA1-EBV-LMP2 fusion proteins, for HLA-B57-restricted epitopes. Using immunoprecipitation, we show that the HLA-B57-restricted peptide A2-LMP2KLRKGA is contained within the EBV-LMP2 protein. Furthermore, by screening a series of in vitro EBV-transformed lymphoblastoid cell lines (LCLs) for T cell reactivity to the A2-LMP2KLRKGA epitope, we show that HLA-B57-restricted epitopes encoded by A2-LMP2 may exist in vivo. A peptide similar to A2-LMP2KLRKGA, B7 (LMP2KLRKGKFV) was found to be shared between EBV and human cytomegalovirus (HCMV) and to contain an A2-restricted motif. Similar to EBV-LMP2, HCMV-LMP2 also contains a polymorphic HLA-A2-restricted epitope (LMP2KLRKGKFV). Using the HLA-A2 transgenic mice, we show that B7, like A2-LMP2KLRKGA, is HLA-A2-restricted and is recognized by A*0201 transgenic T cells. The demonstration that HLA-B57 shares epitopes with other EBV antigens may explain the predisposition to develop EBV-associated disorders in HLA-B57 carriers.Introducing the Apple Watch Series 4
